    Trabalho de Conclusão de Residência apresentado ao Programa de Pós-graduação em Saúde da Família (Residência Multiprofissional), do Instituto Latino-americano de Ciências da Vida e da Natureza da Universidade Federal da Integração Latino Americana, como requisito parcial para obtenção do título de Especialista em Saúde da Família. Trata-se de um estudo ecológico, realizado no município de Foz do Iguaçu-PR, com o objetivo de analisar os efeitos da implantação das medidas restritivas no enfrentamento da COVID-19. Foram analisados decretos publicados no ano de 2020 e no primeiro quadrimestre de 2021. Após análise primária dos decretos, foram criadas nove categorias de medidas restritivas. Posteriormente, foram elaboradas e analisadas as seguintes variáveis: número de casos novos; número de óbitos; taxa de mortalidade; taxa de incidência e letalidade. Os resultados permitiram compreender a implantação das medidas restritivas no município de Foz do IguaçuPR, como ferramenta de redução dos casos, hospitalizações, e, consequentemente, dos óbitos. Foi possível observar a implementação de medidas restritivas em tempo oportuno e o impacto das flexibilizações no aumento dos casos, principalmente nas primeiras semanas epidemiológicas de 2021, em que se observou o maior pico de casos.     Authors: Popov, Vladimir;

    The debate between the US and China about the responsibility for the COVID-19 pandemic raises important questions about the obligations of national governments in the global health care domain. Whereas the US attempts to put the blame on China do not have any serious legal or moral justifications, there is certainly a rationale for the establishment of certain minimal standards in the provision of health care for particular countries. Externalities in the global health care protection are too obvious – costs of underinvestment into the national health care system are borne not only by the country in question, but by the whole world. It is argued that countries should have certain obligations in providing health care services and protecting the population from diseases, especially infectious diseases, similar to the obligations in the framework of the responsibility to protect (R2P) concept that requires countries to protect their citizens from human rights violations. From the point of view of ensuring high life expectancy at a given level of per capita income and spending on health care, China is doing better than many other countries, including the United States that has high per capita income, spends 17% of GDP on health care, but does not provide universal access to health care and lags behind countries with a similar level of economic development in terms of life expectancy (79 years).

    Trabalho de Conclusão de Curso apresentado ao Instituto Latino-Americano de Arte, Cultura e História da Universidade Federal da Integração Latino-Americana, como requisito parcial à obtenção do título de Bacharel em Antropologia – Diversidade Cultural Latino-Americana. As mulheres da terra – que vivem da terra e fazem a terra viver, que têm intensos sentimentos e ações de cuidado pela terra, o território e suas riquezas naturais – sendo elas mulheres dos campos, das florestas ou urbanas, possuem diversidades de relações e conexões com a terra, através do cultivo, da alimentação, da saúde, da espiritualidade, dos conflitos por território, etc. Este trabalho apresentará um panorama antropológico interseccional (englobando aspectos de gênero, raça e classe social) através da análise crítica de materiais bibliográficos, audiovisuais e entrevistas qualitativas realizadas durante a pandemia de COVID-19 de maneira virtual, sobre as pluralidades de agrosabedorias femininas individuais, coletivas ou ancestrais, e sobre as ações de enfrentamento e resistência às violências a que estão expostas através de construções e desconstruções coletivas em movimentos sociais e organizações e da busca cotidiana por uma vida mais ‘sana’ buscando autonomia e soberania para seus corpos-territórios em conexão com a Pachamama, conformando assim a metodologia de etnografia multissituada. O intuito deste trabalho é realçar as vozes das 12 mulheres entrevistadas que compartilham suas sabedorias e analisar como suas perspectivas pessoais se alinham com as diretrizes dos movimentos sociais mais representativos em seus países, propondo também alternativas de vida mais autônomas e ecológicas para curar as mulheres e a terra em Abya Yala.

    Authors: Bundesministerium für Verkehr und digitale Infrastruktur (BMVI);

    Innovationen sind das Fundament eines zukunftsfähigen Mobilitätssystems Im Jahr 2020 haben sich die klima- und wirtschaftspolitischen Rahmenbedingungen für den Verkehrssektor auf Bundes- und europäischer Ebene verändert. Beispiele sind die in Kraft getretenen CO2-Flottengrenzwerte der EU oder die Nationale Wasserstoffstrategie. Geprägt war das Jahr auch von einem starken Markthochlauf der Elektromobilität mit zum Teil dreistelligen Zuwachsraten pro Monat bei den Elektrofahrzeugen und einem 50-prozentigen Zuwachs an Ladepunkten verglichen zum Vorjahreszeitraum. Laut Kraftfahrtbundesamt waren zum 1. Oktober 2020 in Deutschland knapp 222.000 batterieelektrische und 195.000 Plug-in Hybrid-Fahrzeuge im Bestand. Gleichzeitig erfassten die Folgen der COVID-19-Pandemie nahezu alle Bereiche der Mobilitätwirtschaft – von der Luftfahrt und dem öffentlichen Verkehr über die Automobilindustrie bis hin zu Tourismus und Veranstaltungen. Die NPM hebt aufgrund dieser Entwicklungen den Stellenwert von technischen und gesellschaftlichen Innovationen noch einmal explizit hervor. Innovationen sind Garant für die Zukunftsfähigkeit eines Landes. Im Bereich der Mobilität werden Innovationen mehr denn je durch die weiter zunehmenden Klimaschutzanstrengungen und die umfassende Digitalisierung befeuert. Mit resilienten Maßnahmen eine bessere Zukunft vorbereiten Sowohl in Deutschland als auch auf EU-Ebene hat die wirtschaftliche Erholung neben der Eindämmung der Pandemie oberste Priorität. Das zur Krisenbewältigung im Juni 2020 beschlossene Corona-Konjunkturpaket ist an Klimaschutz und der Förderung von Zukunftstechnologien ausgerichtet. Es setzt sich für den öffentlichen Verkehr ein, fördert Wasserstofftechnologien sowie Elektromobilität und unterstützt die Fahrzeugindustrie in ihrer Transformation. In einem Sonderauftrag der Konzertierten Aktion Mobilität legte die NPM Handlungsempfehlungen für einen optimierten elektrischen Nutzungsgrad von Plugin-Hybridfahrzeugen (PHEV) vor. Die Empfehlungen entlang der Themen Fahrzeugtechnik, Lade- und Netzinfrastruktur sowie Nutzungsverhalten zielen darauf ab, dass sie einen elektrischen Fahranteil von PHEV von mindestens 50 % unterstützen. Innovationsmotor Digitalisierung schafft nutzerfreundliche Mobilität Digitalisierung ermöglicht eine plattformbasierte intermodale Vernetzung von Verkehrsmitteln und damit eine Optimierung von Wegeketten. Bürgerinnen und Bürger wünschen sich eine einfache, jederzeit verfügbare, bezahlbare und komfortable Mobilität „aus einer Hand“. Eine wichtige Voraussetzung hierfür ist die Schaffung eines Mobilitätsdatenökosystems, das einen sicheren Datenaustausch zwischen Fahrzeugherstellern, Mobilitätsanbietern und Infrastrukturbereitstellern leistet. Damit können neue Dienstleistungen umgesetzt, automatisierte Fahrfunktionen unterstützt und perspektivisch der Weg zur Nutzung autonomer Flotten geebnet werden. Standards und Normen beschleunigen die Umsetzung nachhaltiger Lösungen Um die Transformation im Mobilitätssektor zu beschleunigen, braucht es Standards und Normen, an denen Unternehmen ihre Entwicklungen ausrichten können. Die NPM hat zum intelligenten Lastmanagement sowie zum automatisierten und vernetzten Fahren Schwerpunkt-Roadmaps vorgelegt, die wichtige Fragen der Sektorkopplung bzw. der Typgenehmigung und Zertifizierung adressieren Darüber hinaus hat sie aufgezeigt, wie Standards und Normen die Marktfähigkeit von Innovationen für eine nachhaltige Mobilität, zum Beispiel über ein einheitliches System der Bilanzierung, fördern können. Für wirksamen Klimaschutz CO2-Ziele in konkrete Maßnahmen übersetzen Um die CO2-Minderungslücke im Verkehrssektor zu schließen, besteht insbesondere im Nutzfahrzeugbereich ein erheblicher Handlungsdruck. Trabalho de Conclusão de Curso apresentado ao Instituto Latino-Americano de Ciências da Vida e da Natureza da Universidade Federal da Integração Latino-Americana, como requisito parcial à obtenção do título de Licenciado em Ciências da Natureza – Biologia, Física e Química. As transformações ocorridas com o avanço do novo coronavírus, causador da doença denominada COVID-19, provocaram diversas mudanças, atingindo várias áreas da sociedade, seja ela econômica, social e, até mesmo, a educacional. E com essas mudanças a consolidação de uma nova modalidade: O ensino Remoto. O ensino remoto trouxe novos desafios e um deles foi fomentar experiências de pesquisa a distância, principalmente no ensino de física, uma vez que existem dificuldades de aprendizagem e muitas vezes a impossibilidade de o aluno relacionar a teoria observada em aula com a realidade que o cerca. O ensino de Física nas escolas se encontra com algumas dificuldades, o que torna cada vez menos atrativo para os jovens a escolha dessa profissão. Tendo em vista que o estudo da física é muito importante, pois coloca os alunos frente a situações concretas e reais, situações essas que os princípios físicos podem responder, ajudando a compreender a natureza nutrindo e alimentando um gosto pela ciência. O uso de novas plataformas e softwares atrai mais atenção dos alunos, fazendo com que eles adquiram maior conhecimento sobre o assunto estudado. Algumas plataformas são muito utilizadas para demonstrar experimentos ou até mesmo quizzes, trazendo certa competitividade saudável aos alunos, gerando conhecimento e interesse pelos temas utilizados. A metodologia utilizada foi a de representação Social, com 2 professores do Colégio Cívico Militar Professora Carmelita Souza Dias, Foz do Iguaçu/PR

    Trata-se de um relato de experiência vivenciada no campo prático durante o internato em Urgência e Emergência do Sistema Único de Saúde, módulo esse necessário para a formação médica. Estão representados em forma descritiva os aprendizados adquiridos baseados na vivencia dos atendimentos prestados nos meses de agosto à dezembro do ano de 2021. As práticas foram em diferentes cenários de aprendizagem em Foz do Iguaçu: Hospital Municipal Padre Germano Lauck e duas Unidades de Pronto Atendimento (Dr. Walter Cavalcante Barbosa e João Samek). Ao todo foram 42 dias e meio de prática divididos em plantões de 12 horas/dia, diurnos e noturnos. Tem como objetivo descrever os casos clínicos vivenciados nos campos de prática selecionados elucidando as discussões com embasamento científico. A metodologia foi relato de caso e foi estruturado conforme orientado no plano de ensino do módulo de urgência e emergência da Universidade Federal da Integração Latino-Americana (UNILA). Ao todo foram selecionados sete temas clínicos estruturado em anamnese, exame físico, exames complementares, diagnósticos diferenciais, hipótese diagnóstica seguida de discussões das condutas clínicas. Justifica-se a escolha destes pela constante ocorrência nos diversos espaços.Ainda contempla a minha vivência pessoal durante o período da pandemia Covid-19. Concluiu se que o internato permite o aluno ampliar os horizontes, adquirir conhecimentos de outras realidades e aprimorar habilidades e competências, entre elas, raciocínio clínico e a organização. Conhecer diferentes locais e equipes possibilitou entender a dinâmica do processo de trabalho do médico e refletir sobre as possíveis soluções e ações para os problemas levantados.

    Am 4./5. Mai 2020 fand der 10. KIM Workshop statt. Durch die COVID-19-Pandemie konnte die Veranstaltung nicht wie gewohnt in Mannheim abgehalten werden, sondern wurde kurzerhand in eine Online-Veranstaltung umgewandelt. Im Folgenden möchten wir auf einige organisatorische Punkte dieser Online-Veranstaltung eingehen und unsere Erfahrungen dabei für andere Online-Veranstalter teilen. Zudem haben wir gegen Ende des Online-Workshops auch einige Fragen an die TeilnehmerInnen gestellt, welche hier zusammen mit den Antworten integriert werden.

    This paper analyzes the moderation effect of government responses on the impact of the COVID-19 pandemic, proxied by the daily growth in COVID-19 cases and deaths, on the capital market, i.e., the S&P 500 firm’s daily returns. Using the Oxford COVID-19 Government Response Tracker, we monitor 16 daily indicators for government actions across the fields of containment and closure, economic support, and health for 180 countries in the period from January 1, 2020 to March 15, 2021. We find that government responses mitigate the negative stock market impact and that investors’ sentiment is sensitive to a firm’s country-specific revenue exposure to COVID-19. Our findings indicate that the mitigation effect is stronger for firms that are highly exposed to COVID-19 on the sales side. In more detail, containment and closure policies and economic support mitigate negative stock market impacts, while health system policies support further declines. For firms with high revenue exposure to COVID-19, the mitigation effect is stronger for government economic support and health system initiatives. Containment and closure policies do not mitigate stock price declines due to growing COVID-19 case numbers. Our results hold even after estimating the spread of the pandemic with an epidemiological standard model, namely, the susceptible-infectious-recovered model.

    Authors: Joppich, Markus;

    This thesis presents new methods for the analysis of high-throughput data from modern sources in the context of complex human diseases, at the example of a bioinformatics analysis workflow. New measurement techniques improve the resolution with which cellular and molecular processes can be monitored. While RNA sequencing (RNA-seq) measures mRNA expression, single-cell RNA-seq (scRNA-seq) resolves this on a per-cell basis. Long-read sequencing is increasingly used in genomics. With imaging mass spectrometry (IMS) the protein level in tissues is measured spatially resolved. All these techniques induce specific challenges, which need to be addressed with new computational methods. Collecting knowledge with contextual annotations is important for integrative data analyses. Such knowledge is available through large literature repositories, from which information, such as miRNA-gene interactions, can be extracted using text mining methods. After aggregating this information in new databases, specific questions can be answered with traceable evidence. The combination of experimental data with these databases offers new possibilities for data integrative methods and for answering questions relevant for complex human diseases. Several data sources are made available, such as literature for text mining miRNA-gene interactions (Chapter 2), next- and third-generation sequencing data for genomics and transcriptomics (Chapters 4.1, 5), and IMS for spatially resolved proteomics (Chapter 4.4). For these data sources new methods for information extraction and pre-processing are developed. For instance, third-generation sequencing runs can be monitored and evaluated using the poreSTAT and sequ-into methods. The integrative (down-stream) analyses make use of these (heterogeneous) data sources. The cPred method (Chapter 4.2) for cell type prediction from scRNA-seq data was successfully applied in the context of the SARS-CoV-2 pandemic. The robust differential expression (DE) analysis pipeline RoDE (Chapter 6.1) contains a large set of methods for (differential) data analysis, reporting and visualization of RNA-seq data. Topics of accessibility of bioinformatics software are discussed along practical applications (Chapter 3). The developed miRNA-gene interaction database gives valuable insights into atherosclerosis-relevant processes and serves as regulatory network for the prediction of active miRNA regulators in RoDE (Chapter 6.1). The cPred predictions, RoDE results, scRNA-seq and IMS data are unified as input for the 3D-index Aorta3D (Chapter 6.2), which makes atherosclerosis related datasets browsable. Finally, the scRNA-seq analysis with subsequent cPred cell type prediction, and the robust analysis of bulk-RNA-seq datasets, led to novel insights into COVID-19. Taken all discussed methods together, the integrative analysis methods for complex human disease contexts have been improved at essential positions. Die Dissertation beschreibt Methoden zur Prozessierung von aktuellen Hochdurchsatzdaten, sowie Verfahren zu deren weiterer integrativen Analyse. Diese findet Anwendung vor allem im Kontext von komplexen menschlichen Krankheiten. Neue Messtechniken erlauben eine detailliertere Beobachtung biomedizinischer Prozesse. Mit RNA-Sequenzierung (RNA-seq) wird mRNA-Expression gemessen, mit Hilfe von moderner single-cell-RNA-seq (scRNA-seq) sogar für (sehr viele) einzelne Zellen. Long-Read-Sequenzierung wird zunehmend zur Sequenzierung ganzer Genome eingesetzt. Mittels bildgebender Massenspektrometrie (IMS) können Proteine in Geweben räumlich aufgelöst quantifiziert werden. Diese Techniken bringen spezifische Herausforderungen mit sich, die mit neuen bioinformatischen Methoden angegangen werden müssen. Für die integrative Datenanalyse ist auch die Gewinnung von geeignetem Kontextwissen wichtig. Wissenschaftliche Erkenntnisse werden in Artikeln veröffentlicht, die über große Literaturdatenbanken zugänglich sind. Mittels Textmining können daraus Informationen extrahiert werden, z.B. miRNA-Gen-Interaktionen, die in eigenen Datenbank aggregiert werden um spezifische Fragen mit nachvollziehbaren Belegen zu beantworten. In Kombination mit experimentellen Daten bieten sich so neue Möglichkeiten für integrative Methoden. Durch die Extraktion von Rohdaten und deren Vorprozessierung werden mehrere Datenquellen erschlossen, wie z.B. Literatur für Textmining von miRNA-Gen-Interaktionen (Kapitel 2), Long-Read- und RNA-seq-Daten für Genomics und Transcriptomics (Kapitel 4.2, 5) und IMS für Protein-Messungen (Kapitel 4.4). So dienen z.B. die poreSTAT und sequ-into Methoden der Vorprozessierung und Auswertung von Long-Read-Sequenzierungen. In der integrativen (down-stream) Analyse werden diese (heterogenen) Datenquellen verwendet. Für die Bestimmung von Zelltypen in scRNA-seq-Experimenten wurde die cPred-Methode (Kapitel 4.2) erfolgreich im Kontext der SARS-CoV-2-Pandemie eingesetzt. Auch die robuste Pipeline RoDE fand dort Anwendung, die viele Methoden zur (differentiellen) Datenanalyse, zum Reporting und zur Visualisierung bereitstellt (Kapitel 6.1). Themen der Benutzbarkeit von (bioinformatischer) Software werden an Hand von praktischen Anwendungen diskutiert (Kapitel 3). Die entwickelte miRNA-Gen-Interaktionsdatenbank gibt wertvolle Einblicke in Atherosklerose-relevante Prozesse und dient als regulatorisches Netzwerk für die Vorhersage von aktiven miRNA-Regulatoren in RoDE (Kapitel 6.1). Die cPred-Methode, RoDE-Ergebnisse, scRNA-seq- und IMS-Daten werden im 3D-Index Aorta3D (Kapitel 6.2) zusammengeführt, der relevante Datensätze durchsuchbar macht. Die diskutierten Methoden führen zu erheblichen Verbesserungen für die integrative Datenanalyse in komplexen menschlichen Krankheitskontexten.

    Authors: Popov, Vladimir;

    Authors: Bundesministerium für Verkehr und digitale Infrastruktur (BMVI);

    Innovationen sind das Fundament eines zukunftsfähigen Mobilitätssystems Im Jahr 2020 haben sich die klima- und wirtschaftspolitischen Rahmenbedingungen für den Verkehrssektor auf Bundes- und europäischer Ebene verändert. Beispiele sind die in Kraft getretenen CO2-Flottengrenzwerte der EU oder die Nationale Wasserstoffstrategie. Geprägt war das Jahr auch von einem starken Markthochlauf der Elektromobilität mit zum Teil dreistelligen Zuwachsraten pro Monat bei den Elektrofahrzeugen und einem 50-prozentigen Zuwachs an Ladepunkten verglichen zum Vorjahreszeitraum. Laut Kraftfahrtbundesamt waren zum 1. Oktober 2020 in Deutschland knapp 222.000 batterieelektrische und 195.000 Plug-in Hybrid-Fahrzeuge im Bestand. Gleichzeitig erfassten die Folgen der COVID-19-Pandemie nahezu alle Bereiche der Mobilitätwirtschaft – von der Luftfahrt und dem öffentlichen Verkehr über die Automobilindustrie bis hin zu Tourismus und Veranstaltungen. Die NPM hebt aufgrund dieser Entwicklungen den Stellenwert von technischen und gesellschaftlichen Innovationen noch einmal explizit hervor. Innovationen sind Garant für die Zukunftsfähigkeit eines Landes. Im Bereich der Mobilität werden Innovationen mehr denn je durch die weiter zunehmenden Klimaschutzanstrengungen und die umfassende Digitalisierung befeuert. Mit resilienten Maßnahmen eine bessere Zukunft vorbereiten Sowohl in Deutschland als auch auf EU-Ebene hat die wirtschaftliche Erholung neben der Eindämmung der Pandemie oberste Priorität. Das zur Krisenbewältigung im Juni 2020 beschlossene Corona-Konjunkturpaket ist an Klimaschutz und der Förderung von Zukunftstechnologien ausgerichtet. Es setzt sich für den öffentlichen Verkehr ein, fördert Wasserstofftechnologien sowie Elektromobilität und unterstützt die Fahrzeugindustrie in ihrer Transformation. In einem Sonderauftrag der Konzertierten Aktion Mobilität legte die NPM Handlungsempfehlungen für einen optimierten elektrischen Nutzungsgrad von Plugin-Hybridfahrzeugen (PHEV) vor. Die Empfehlungen entlang der Themen Fahrzeugtechnik, Lade- und Netzinfrastruktur sowie Nutzungsverhalten zielen darauf ab, dass sie einen elektrischen Fahranteil von PHEV von mindestens 50 % unterstützen. Innovationsmotor Digitalisierung schafft nutzerfreundliche Mobilität Digitalisierung ermöglicht eine plattformbasierte intermodale Vernetzung von Verkehrsmitteln und damit eine Optimierung von Wegeketten. Bürgerinnen und Bürger wünschen sich eine einfache, jederzeit verfügbare, bezahlbare und komfortable Mobilität „aus einer Hand“. Eine wichtige Voraussetzung hierfür ist die Schaffung eines Mobilitätsdatenökosystems, das einen sicheren Datenaustausch zwischen Fahrzeugherstellern, Mobilitätsanbietern und Infrastrukturbereitstellern leistet. Damit können neue Dienstleistungen umgesetzt, automatisierte Fahrfunktionen unterstützt und perspektivisch der Weg zur Nutzung autonomer Flotten geebnet werden. Standards und Normen beschleunigen die Umsetzung nachhaltiger Lösungen Um die Transformation im Mobilitätssektor zu beschleunigen, braucht es Standards und Normen, an denen Unternehmen ihre Entwicklungen ausrichten können. Die NPM hat zum intelligenten Lastmanagement sowie zum automatisierten und vernetzten Fahren Schwerpunkt-Roadmaps vorgelegt, die wichtige Fragen der Sektorkopplung bzw. der Typgenehmigung und Zertifizierung adressieren Darüber hinaus hat sie aufgezeigt, wie Standards und Normen die Marktfähigkeit von Innovationen für eine nachhaltige Mobilität, zum Beispiel über ein einheitliches System der Bilanzierung, fördern können. Für wirksamen Klimaschutz CO2-Ziele in konkrete Maßnahmen übersetzen Um die CO2-Minderungslücke im Verkehrssektor zu schließen, besteht insbesondere im Nutzfahrzeugbereich ein erheblicher Handlungsdruck. Trabalho de Conclusão de Curso apresentado ao Instituto Latino-Americano de Ciências da Vida e da Natureza da Universidade Federal da Integração Latino-Americana, como requisito parcial à obtenção do título de Licenciado em Ciências da Natureza – Biologia, Física e Química. As transformações ocorridas com o avanço do novo coronavírus, causador da doença denominada COVID-19, provocaram diversas mudanças, atingindo várias áreas da sociedade, seja ela econômica, social e, até mesmo, a educacional. E com essas mudanças a consolidação de uma nova modalidade: O ensino Remoto. O ensino remoto trouxe novos desafios e um deles foi fomentar experiências de pesquisa a distância, principalmente no ensino de física, uma vez que existem dificuldades de aprendizagem e muitas vezes a impossibilidade de o aluno relacionar a teoria observada em aula com a realidade que o cerca. O ensino de Física nas escolas se encontra com algumas dificuldades, o que torna cada vez menos atrativo para os jovens a escolha dessa profissão. Tendo em vista que o estudo da física é muito importante, pois coloca os alunos frente a situações concretas e reais, situações essas que os princípios físicos podem responder, ajudando a compreender a natureza nutrindo e alimentando um gosto pela ciência. O uso de novas plataformas e softwares atrai mais atenção dos alunos, fazendo com que eles adquiram maior conhecimento sobre o assunto estudado. Algumas plataformas são muito utilizadas para demonstrar experimentos ou até mesmo quizzes, trazendo certa competitividade saudável aos alunos, gerando conhecimento e interesse pelos temas utilizados. A metodologia utilizada foi a de representação Social, com 2 professores do Colégio Cívico Militar Professora Carmelita Souza Dias, Foz do Iguaçu/PR

    Authors: Joppich, Markus;

    This thesis presents new methods for the analysis of high-throughput data from modern sources in the context of complex human diseases, at the example of a bioinformatics analysis workflow. New measurement techniques improve the resolution with which cellular and molecular processes can be monitored. While RNA sequencing (RNA-seq) measures mRNA expression, single-cell RNA-seq (scRNA-seq) resolves this on a per-cell basis. Long-read sequencing is increasingly used in genomics. With imaging mass spectrometry (IMS) the protein level in tissues is measured spatially resolved. All these techniques induce specific challenges, which need to be addressed with new computational methods. Collecting knowledge with contextual annotations is important for integrative data analyses. Such knowledge is available through large literature repositories, from which information, such as miRNA-gene interactions, can be extracted using text mining methods. After aggregating this information in new databases, specific questions can be answered with traceable evidence. The combination of experimental data with these databases offers new possibilities for data integrative methods and for answering questions relevant for complex human diseases. Several data sources are made available, such as literature for text mining miRNA-gene interactions (Chapter 2), next- and third-generation sequencing data for genomics and transcriptomics (Chapters 4.1, 5), and IMS for spatially resolved proteomics (Chapter 4.4). For these data sources new methods for information extraction and pre-processing are developed. For instance, third-generation sequencing runs can be monitored and evaluated using the poreSTAT and sequ-into methods. The integrative (down-stream) analyses make use of these (heterogeneous) data sources. The cPred method (Chapter 4.2) for cell type prediction from scRNA-seq data was successfully applied in the context of the SARS-CoV-2 pandemic. The robust differential expression (DE) analysis pipeline RoDE (Chapter 6.1) contains a large set of methods for (differential) data analysis, reporting and visualization of RNA-seq data. Topics of accessibility of bioinformatics software are discussed along practical applications (Chapter 3). The developed miRNA-gene interaction database gives valuable insights into atherosclerosis-relevant processes and serves as regulatory network for the prediction of active miRNA regulators in RoDE (Chapter 6.1). The cPred predictions, RoDE results, scRNA-seq and IMS data are unified as input for the 3D-index Aorta3D (Chapter 6.2), which makes atherosclerosis related datasets browsable. Finally, the scRNA-seq analysis with subsequent cPred cell type prediction, and the robust analysis of bulk-RNA-seq datasets, led to novel insights into COVID-19. Taken all discussed methods together, the integrative analysis methods for complex human disease contexts have been improved at essential positions. Die Dissertation beschreibt Methoden zur Prozessierung von aktuellen Hochdurchsatzdaten, sowie Verfahren zu deren weiterer integrativen Analyse. Diese findet Anwendung vor allem im Kontext von komplexen menschlichen Krankheiten. Neue Messtechniken erlauben eine detailliertere Beobachtung biomedizinischer Prozesse. Mit RNA-Sequenzierung (RNA-seq) wird mRNA-Expression gemessen, mit Hilfe von moderner single-cell-RNA-seq (scRNA-seq) sogar für (sehr viele) einzelne Zellen. Long-Read-Sequenzierung wird zunehmend zur Sequenzierung ganzer Genome eingesetzt. Mittels bildgebender Massenspektrometrie (IMS) können Proteine in Geweben räumlich aufgelöst quantifiziert werden. Diese Techniken bringen spezifische Herausforderungen mit sich, die mit neuen bioinformatischen Methoden angegangen werden müssen. Für die integrative Datenanalyse ist auch die Gewinnung von geeignetem Kontextwissen wichtig. Wissenschaftliche Erkenntnisse werden in Artikeln veröffentlicht, die über große Literaturdatenbanken zugänglich sind. Mittels Textmining können daraus Informationen extrahiert werden, z.B. miRNA-Gen-Interaktionen, die in eigenen Datenbank aggregiert werden um spezifische Fragen mit nachvollziehbaren Belegen zu beantworten. In Kombination mit experimentellen Daten bieten sich so neue Möglichkeiten für integrative Methoden. Durch die Extraktion von Rohdaten und deren Vorprozessierung werden mehrere Datenquellen erschlossen, wie z.B. Literatur für Textmining von miRNA-Gen-Interaktionen (Kapitel 2), Long-Read- und RNA-seq-Daten für Genomics und Transcriptomics (Kapitel 4.2, 5) und IMS für Protein-Messungen (Kapitel 4.4). So dienen z.B. die poreSTAT und sequ-into Methoden der Vorprozessierung und Auswertung von Long-Read-Sequenzierungen. In der integrativen (down-stream) Analyse werden diese (heterogenen) Datenquellen verwendet. Für die Bestimmung von Zelltypen in scRNA-seq-Experimenten wurde die cPred-Methode (Kapitel 4.2) erfolgreich im Kontext der SARS-CoV-2-Pandemie eingesetzt. Auch die robuste Pipeline RoDE fand dort Anwendung, die viele Methoden zur (differentiellen) Datenanalyse, zum Reporting und zur Visualisierung bereitstellt (Kapitel 6.1). Themen der Benutzbarkeit von (bioinformatischer) Software werden an Hand von praktischen Anwendungen diskutiert (Kapitel 3). Die entwickelte miRNA-Gen-Interaktionsdatenbank gibt wertvolle Einblicke in Atherosklerose-relevante Prozesse und dient als regulatorisches Netzwerk für die Vorhersage von aktiven miRNA-Regulatoren in RoDE (Kapitel 6.1). Die cPred-Methode, RoDE-Ergebnisse, scRNA-seq- und IMS-Daten werden im 3D-Index Aorta3D (Kapitel 6.2) zusammengeführt, der relevante Datensätze durchsuchbar macht. Die diskutierten Methoden führen zu erheblichen Verbesserungen für die integrative Datenanalyse in komplexen menschlichen Krankheitskontexten.
